以西藏自治区拉萨市尼木县产的藏香柏嫩枝为原料,采用水蒸气蒸馏法制备藏香柏精油,在单因素实验基础上,采用响应面法优化提取工艺,采用气相色谱-质谱联用技术分析精油的化学成分.结果表明:在料液比1:16 g·mL-1、浸泡时间2.76 h、温度190℃的条件下藏香柏精油的出油率最高;精油的主要成分为乙酸龙脑酯、石竹素、α-萜品油烯、β-古巴烯、松香醛、柠檬烯及γ-桉叶醇等.研究证明,响应面法优化藏香柏精油提取工艺的路径可行,该研究结果对深入开发藏香柏精油药用功效具有借鉴意义.
Abstract:Based on single-factor experiments,we extracted the essential oil from Tibetan cedar produced in Nyemo County,Lhasa City,Tibet Autonomous Region,with steam distillation technology and optimized the extraction process by response surface methodology.Meanwhile,we identified the chemical composition in the essential oil by gas chromatography-mass spectrometry technology.The results showed that the highest yield of the essential oil was acquired when liquid to material ratio was 1∶16 g·mL-1,soaking time was 2.76 h,and extraction temperature was 190 ℃.The main components of the essential oil included bornyl acetate,caryophyllene,α-terpinene,β-copaene,rosin aldehyde,limonene,and γ-eucalyptol,etc.The above results prove that response surface methodology is a feasible way to optimize the extraction process of the essential oil in Tibetan cedar,which is of great significance to explore the medicinal function of the essential oil.
