采用高效液相色谱法对生普洱与熟普洱茶中咖啡因的含量进行测定.色谱柱为Kromasil C18;流动相:水-甲醇-乙酸-二甲基酰胺(70∶30∶0.05∶0.25,V/V);色谱柱温:25℃;进样量:5μL;检测器:紫外检测器;扫描波长:210~400 nm;检测波长:274 nm.方法变异系数小于3.96%,回收率为90.62%~103.8%.生普洱茶和熟普洱茶咖啡因含量分别为19.92 mg/g和28.58 mg/g.
Abstract:The content of caffeine in raw and cooked puer tea were determined by HPLC,with Kromasil C18 as Column,methanol-water-acetic acid-dimethyl amide( 70 ∶ 30 ∶ 0.05 ∶ 0.05,V / V) as mobile phase,25 ℃ as column temperature,274 nm as the detection wavelength,and 5μL as the injection volume. The coefficient of variation in this paper is less than 3.96%,and the recovery is 90.62% ~ 90.62%.The content of caffeine in raw and cooked puer tea were 19.92 mg / g and 28.58 mg / g respectively.
